Acuitas Therapeutics Inc. has quietly become one of Canada’s most lucrative biopharma companies by providing a critical piece of technology
used to deliver the Pfizer-BioNTech COVID-19 vaccine inside the body. Now, the Vancouver company has inked a deal with Pfizer Inc. to expand
its efforts to help deliver a range of other treatments.

The U.S. drug giant on Monday announced an option deal to use Acuitas’s lipid nanoparticle (LNP) technology to develop up to 10 vaccines
and therapies, after LNP proved to be a critical part of the development of its COVID-19 vaccine with BioNTech SE. Terms were not disclosed.
The collaboration was one of a set of partnership deals unveiled by
Pfizer to expand its use of messenger RNA (mRNA) technology.

It’s the first direct engagement between Pfizer and Acuitas, which had worked with BioNTech before the German company
joined with Pfizer to produce their mRNA vaccine. And it points to more opportunities for Acuitas, which has grown by about
75 per cent annually in recent years
and whose revenues exceed US$100-million, with most of that falling to the bottom line.

New study: 133x risk of myocarditis after COVID vaccination
Comparisons with myocarditis rates following infection now irrelevant as vaccination no longer prevents infection.
A recent study published on January 25, 2022, on JAMA Network, has shown that the risk of myocarditis following mRNA COVID vaccination is around
133 times greater than the background risk in the population.

The study, conducted by researchers from the U.S. Centers for Disease Control (CDC) as well as from several U.S. universities and hospitals,
examined the effects of vaccination with products manufactured by Pfizer-BioNTech and Moderna. The study’s authors used data obtained from the
CDC’s VAERS reporting system which were cross-checked to ensure they complied with CDC’s definition of myocarditis; they also noted that given the
passive nature of the VAERS system, the number of
reported incidents is likely to be an underestimate of the extent of the phenomenon.

1626 cases of myocarditis were studied, and the results showed that the Pfizer-BioNTech product was most associated with higher risk, with 105.9
cases per million doses after the second vaccine shot in the 16 to 17 age group for males, and 70.7 cases per million doses after the second shot in
the 12 to 15 age group for males. The 18 to 24 male age group also saw significantly higher rates of myocarditis for both Pfizer’s and Moderna’s products
(52.4 and 56.3 cases per million respectively).

Abstract

The movement of proteins between the cytoplasm and nucleus mediated by the importin superfamily of proteins is essential to many cellular processes,
including differentiation and development, and is
critical to disease states such as viral disease and oncogenesis. We recently developed a high-throughput screen to identify specific and general inhibitors
of protein nuclear import, from which ivermectin was identified as a potential inhibitor of importin α/β-mediated transport. In the present study, we
characterized in detail the nuclear transport inhibitory properties of ivermectin, demonstrating that it is a broad-spectrum inhibitor of importin α/β
nuclear import, with no effect on a range of other nuclear import pathways, including that mediated by importin β1 alone. Importantly, we establish
for the first time that ivermectin has potent antiviral activity towards both HIV-1 and dengue virus, both of which are strongly reliant on importin α/β
nuclear import, with respect to the HIV-1 integrase and NS5 (non-structural protein 5) polymerase proteins respectively. Ivermectin would appear to be
an invaluable tool for the study of protein nuclear import, as well as the basis for future development of antiviral agents.

Abstract
Constitutive activation of canonical WNT-TCF signaling is implicated in multiple diseases, including intestine and lung cancers, but there are no
WNT-TCF antagonists in clinical use. We have performed a repositioning screen for WNT-TCF response blockers aiming to recapitulate the genetic
blockade afforded by dominant-negative TCF. We report that Ivermectin inhibits the expression of WNT-TCF targets, mimicking dnTCF, and that its
low concentration effects are rescued by direct activation by TCFVP16. Ivermectin inhibits the proliferation and increases apoptosis of various
human cancer types. It represses the levels of C-terminal β-CATENIN phosphoforms and of CYCLIN D1 in an okadaic acid-sensitive manner,
indicating its action involves protein phosphatases. In vivo, Ivermectin selectively inhibits TCF-dependent, but not TCF-independent, xenograft
growth without obvious side effects. Analysis of single semi-synthetic derivatives highlights Selamectin, urging its clinical testing and the
exploration of the macrocyclic lactone chemical space. Given that Ivermectin is a safe anti-parasitic agent used by > 200 million people against
river blindness, our results suggest its additional use as a therapeutic WNT-TCF pathway response blocker to treat WNT-TCF-dependent diseases
including multiple cancers.
