What does “born again” actually mean? Why did Jesus use it when talking to Nicodemus?

Jesus told Nicodemus, a representative of the nation of Israel, they must be “born again”. When was the first birth? The first birth happened when the Israelites passed through the parted waters while escaping Egypt. Why does Jesus reference the Old Testament and Moses’ staff?
