This video presents a PhD research work on the potential anticancerous of the Graviola fruit (Soursop) extract using in vitro method to assess the antiproliferative effect of the Graviola fruit on breast and colon cancer cell lines and assess in vivo the acute toxicity of the fruit extract on zebrafish model. The research started with the extraction and optimization of extraction conditions of the ionic liquid-based microwave-assisted extraction using Design of Experiment software to maximize the yield and minimize the IC50 of the Graviola fruit extract on breast MCF-7 and colon HT29 cancer cells. Then the crude extract of ionic liquid-Graviola fruit extract (IL-GFE) was fractionated using Thin Layer Chromatography and Column Chromatography. After that, the antiproliferative effect of the obtained extract on MCF-7, HT29 cancer cells and normal VERO cell lines was evaluated using MTT assay. Additionally, the acute toxicity of IL-GFE on zebrafish embryos and larvae was assessed. Then, the cell cycle phase distribution and apoptosis/necrosis induction in the treated cancer cells with crude IL-GFE was determined via flow cytometry technique. Finally, the effect of the crude IL-GFE on the metabolome of human adenocarcinoma MCF-7 and HT-29 cancer cell lines using GC-TOFMS-based metabolic profiling and the chemometric method was studied using XLSTAT software and KEGG pathways database and Ingenuity Pathway Analysis (IPA).
