Esketamine is a drug that is used to treat severe depression that does not respond to other treatments. It is a nasal spray that is derived from ketamine, an anesthetic and hallucinogen.

Esketamine works by blocking a receptor in the brain called NMDA, which may affect mood and cognition. Esketamine was approved by the FDA in 2019 and is sold under the brand name Spravato.

Esketamine must be used with another antidepressant and under medical supervision, as it can cause side effects such as dissociation, dizziness, nausea, and increased blood pressure.

Because of the risks with this medication, esketamine is available only through a special restricted distribution program. A program called Spravato Risk Evaluation and Mitigation Strategies (REMS) program.

You, your doctor, and your pharmacy must be enrolled in the Spravato REMS program before you can receive this medication.

You will use esketamine nasal spray in a medical facility under the observation of a doctor or other healthcare professional.
