Trimethylglycine is an amino acid that has been used as a traditional remedy in Southeast Asia for hepatic disorders. Rich dietary sources of this amino acid include seafood, spinach, wheat germ and wheat brain. Trimethylglycine has been known to play an essential role in the health of the cardiovascular system, liver function, amino acid metabolism and glucose metabolism. Accumulating evidence reports that it suppresses pro inflammatory signaling during aging. This amino acid has been found to suppress nuclear factor kappa B and its related gene expressions of cyclooxygenase 2. Nuclear factor kappa B or NF-κB is a family of transcription factors that exhibit an essential role in inflammation associated with cell senescence and tissue aging. NF-κB activation is increased in a variety of organs with aging and is associated with age related degenerative diseases including Alzheimer's disease. Indeed, the suppression of NF-κB pathway by trimethylglycine has been proposed to delay the aging process and the progression of age related diseases. Trimethylglycine has also been reported to inhibit the leucin rich family, pyrin containing 4 or NLRP3 inflammasome related proteins, which are involved in initiating inflammaging. Inflammaging is a chronic low grade inflammation, associated with age related diseases. Inhibition of the NLRP3 inflammasome pathway by trimethylglycine enhances the ability of trimethylglycine to delay the aging process and prevent ailments associated with aging. The recommended daily intake of trimethylglycine is 9 to 15 grams for human and distributes primarily to the kidney, liver, and brain.
