Mitochondrial derived peptides or MDP's are series of bioactive peptides hidden in the mitochondrial DNA. MDP's are widely found in the heart, vascular wall, kidney, skeletal muscle, and colon. These peptides may influence the expression of nuclear genes that maintain mitochondrial function and play protective roles against chronic and age-related diseases. The first putative set of mitochondrial-derived peptides is humanin. Humanin has been demonstrated in vitro in human cells and in vivo in aged mice to exhibit strong neuroprotective effect in response to conditions of stress. In a study, human neuroblastoma cell line was treated with soluble A-β 1-42 with and without humanin analogue. It has been found that humanin analogue against A beta induced reduction of cell metabolism and viability, Furthermore, the protective effects of reactive oxygen species induced by A-β directly in mitochondria. Humanin doesn't only provide neuroprotection against Alzheimer's disease. It also prevents overall cognitive decline associated with aging. Humanin has been demonstrated to be locally released by astroglial cells in the neighbouring synapse, thereby regulating synaptic plasticity in pathophysiological conditions. Humanin also induces the expression of chaperone-mediated autophagy by increasing the substrate binding and translocation into lysosomes, thereby contributing to cellular quality control and maintenance of cellular energy balance. The activity of chaperone-mediated autophagy and humanin decline with age, which contribute to the pathogenesis of age-related diseases such as neurodegeneration and metabolic disease.
